Molecular cloning and isolation of a recombinant alpha-Momorcharin in E. coli against Pyricularia oryzae

Introduction: Alpha-Momorcharin (α-MMC) is a member of the ribosome-inactivating protein (RIP) family that has been widely used as an antitumor, antiviral and antifungal agent. Methods: In this study, codons DNA encoding α-MMC were optimized for expression in E. coli cloned into pET-28a(+) vector. The was then expressed strain BL21 (DE3) purified by nickel affinity chromatography. Results: Under IPTG induction, at approximately 50% total protein, showing high-level recombinant coli. A high amount (70 mg) isolated from 1 L LB culture medium with 95% purity. Interestingly, inhibited mycelial growth Pyricularia oryzae concentration-dependent manner. Conclusion: Using microbial system provides promising method design new agent against pathogens.
